Protocol ItemGuidance
Standard Dose Range100–500 mcg per dose
Higher Range Noted500–1500 mcg per dose has been observed in research with positive results and is often seen in the bio-hacking community
Frequency1–7× per week
Cycle6 weeks on / 6 weeks off
Modification NotesLimited research exists on dosing/cycling schemes for modified Semax versions (N-Acetyl and Amidate). Current dosing/cycling schemes are similar to the original compound.
Conservative StartOften recommended to start with a more conservative dosing/cycling scheme when researching more potent compounds.

What Is It?

Semax

ACTH-derived peptide studied in cognition research.

Bacteriostatic Water

Sterile water containing a bacteriostatic preservative, commonly used when preparing multi-use research vials.

How To Mix Semax

1
Clean

Use alcohol swabs to clean the tops of both vials.

2
Draw BAC Water

Draw the selected amount of bacteriostatic water.

3
Inject Slowly

Add the liquid slowly down the side of the vial.

4
Swirl Gently

Do not shake. Swirl gently until dissolved.

5
Store Properly

Store as directed and protect from heat and light.

Best Practices & Common Mistakes

Best Practices

  • Use sterile technique.
  • Protect from light and heat.
  • Store refrigerated when appropriate.
  • Use clean syringe-unit math before measuring.

Common Mistakes

  • Confusing milligrams with milliliters.
  • Choosing an option with awkward decimal units.
  • Using too little liquid for very small measurements.
  • Shaking the vial aggressively.

Semax Storage & Handling

Lyophilized Powder: −20°C (−4°F) for long-term storage (up to 24 months). Refrigeration 2–8°C (36–46°F) for short-term use (up to ~3 months). Original sealed vial in the freezer is safest.
Reconstituted Solution: 2–8°C (36–46°F), use within ~7–14 days. Keep sealed, avoid light, and do not repeat freeze-thaw cycles.

Frequently Asked Questions

Semax is a synthetic seven-amino-acid peptide (Met-Glu-His-Phe-Pro-Gly-Pro) developed by the Russian Academy of Sciences from a fragment of ACTH, with a Pro-Gly-Pro tail added for stability. It's a nootropic and neuroprotective peptide — used to support focus, memory, and brain recovery — and importantly it was designed to keep the brain effects of ACTH while dropping the hormonal part, so it doesn't raise cortisol. It's most often used as a nasal spray, and also comes as a research powder.
The three are the same core peptide with escalating modifications. Plain Semax has the most clinical data but the shortest action; N-Acetyl Semax (NA-Semax) adds an acetyl group for roughly 2–3× the potency and longer duration; and N-Acetyl Semax Amidate (NASA) adds amidation too, making it the strongest and longest-acting (about 3–5×). That's why the modified versions are dosed at a fraction of standard Semax — matching them one-to-one would be an overdose.
Semax's main documented action is raising BDNF and NGF — growth factors tied to learning, memory, and neuron repair — while modulating dopamine and serotonin, which fits its reported effects on focus, mood, and mental stamina, especially under stress. It is not a classical stimulant and doesn't work like caffeine or amphetamine; effects are most noticeable when the brain is under load. Most evidence is Russian clinical and animal research with limited Western replication, and claims like hair-loss benefit are anecdotal, not established.
Semax is usually used as a pre-made nasal spray, which isn't reconstituted; only a research-grade powder is mixed. For a 10 mg vial, adding 2 mL of bacteriostatic water gives 5 mg/mL, so a 300 mcg amount draws to 0.06 mL (6 units on a U-100 syringe) — and note that N-Acetyl variants need a much smaller amount for the same effect. Because the plasma half-life is only minutes, the peptide clears fast even though its BDNF effect lasts far longer.
Semax is not FDA-approved in the US, though it's a registered prescription medicine in Russia and Ukraine; in the US it's available through 503A compounding pharmacies with a prescription (it was briefly on the FDA's Category 2 compounding list from 2023 until April 2026). Russian trials report a benign profile — mainly transient nasal irritation, mild headache, or a metallic taste — but there's no Western safety oversight or drug-interaction data, and the potent N-Acetyl variants warrant extra caution. Storage: keep the dry powder cold, and refrigerate reconstituted solution or nasal spray at around 4 °C, using it within a few weeks.
Practical takeaway: If your real goal is weight or metabolic health, the most useful next step is discussing approved treatment options with a clinician rather than relying on an unapproved compound.
